An in vitro hatching technique for the eggs of Dicrocoelium dendriticum

Introduction
Private institutions require Dicrocoelium miracidium for their research, the most uses are for the studies of life cycle this trematod, through experimental infection of terrestrial pulmonate or to follow the quantitative variations of different biological parameters in miracidium, Therefore, hatching egg of dicrocoelium is essential in laboratory conditions.The naturally occurring hatching factors of D. dendricum eggs ingested by their terrestrial pulmonate intermediate hosts are essentially unknown.
The present investigation reports on hatching experiments performed with sodium hypochlorite (6-14% active chlorite, M.W; 44-74g/mol).
Materials and methods
D. dendricum eggs were obtained from gall bladders of infected sheep or removed from adult flukes and normally stored in water at 4°C. A 0.5-ml suspension of eggs was transferred into a glass watch. After 30 min, the supernatant was removed, and then, 0.5 ml sodium hypochlorite (0.3% and 0.5% sodium hypochlorite in distilled water) was added to the suspension and left for 10-70 min at room temperature (25–27°C).Then, the hatching rate was calculated under a light microscope.
Results & Conclusion
The results obtained from the hatching rate of Dicrocoelium eggs by 3% in 70 min and 5% in 30 min sodium hypochlorite solutions.
